Hesperetin as a Potential Drug Candidate for the Treatment of Pancreatic Cancer

Abstract Pancreatic cancer remains one of the most lethal malignancies worldwide, characterized by late diagnosis, rapid progression, and poor responsiveness to existing therapeutic modalities. The search for novel, safe, and effective anticancer agents has drawn considerable attention to bioactive natural compounds, particularly flavonoids. Hesperetin, a flavanone derived from citrus fruits, has demonstrated promising anticancer properties in various in vitro and in vivo studies. This mini-review highlights the pharmacological potential of hesperetin against pancreatic cancer, focusing on its physicochemical and pharmacokinetic profiles, biological activities, and underlying mechanisms of action. Hesperetin exerts anti-proliferative, pro-apoptotic, anti-angiogenic, and anti-metastatic effects through modulation of key oncogenic pathways, including PI3K/Akt/mTOR and NF-κB signaling. However, its clinical applicability is limited by poor water solubility, low bioavailability, and rapid metabolic degradation. Advanced nanocarrier-based formulations have been proposed to overcome these limitations and enhance therapeutic efficacy. This review also discusses current challenges and outlines future research directions necessary for the clinical development of hesperetin as a potential therapeutic or adjuvant agent in pancreatic cancer treatment.
